Prokineticin receptors in GtoPdb v.2023.1
نویسندگان
چکیده
Prokineticin receptors, PKR1 and PKR2 (provisional nomenclature as recommended by NC-IUPHAR [26]) respond to the cysteine-rich 81-86 amino-acid peptides prokineticin-1 (also known endocrine gland-derived vascular endothelial growth factor, mambakine) prokineticin-2 (protein Bv8 homologue). An orthologue of PROK1 from black mamba (Dendroaspis polylepis) venom, intestinal toxin 1 (MIT1, [71]) is a potent, non-selective agonist at prokineticin receptors [46], while Bv8, an PROK2 amphibians (Bombina sp., [49]), equipotent recombinant [53], has high potency in macrophage chemotaxis assays, which are lost PKR1-null mice.

Divergent roles of prokineticin receptors in the endothelial cells: angiogenesis and fenestration.
Prokineticins are secreted peptides that activate two G protein-coupled receptors: PKR1 and PKR2. Prokineticins induce angiogenesis and fenestration, but the cognate receptors involved in these functions are unknown. We hypothesized a role for prokineticin receptor signaling pathways and expression profiles in determining the selective effects of prokineticins on coronary endothelial cells (H5V...
متن کاملModeling of Human Prokineticin Receptors: Interactions with Novel Small-Molecule Binders and Potential Off-Target Drugs
BACKGROUND AND MOTIVATION The Prokineticin receptor (PKR) 1 and 2 subtypes are novel members of family A GPCRs, which exhibit an unusually high degree of sequence similarity. Prokineticins (PKs), their cognate ligands, are small secreted proteins of ∼80 amino acids; however, non-peptidic low-molecular weight antagonists have also been identified. PKs and their receptors play important roles und...
متن کاملKallmann Syndrome: Mutations in the Genes Encoding Prokineticin-2 and Prokineticin Receptor-2
Kallmann syndrome combines anosmia, related to defective olfactory bulb morphogenesis, and hypogonadism due to gonadotropin-releasing hormone deficiency. Loss-of-function mutations in KAL1 and FGFR1 underlie the X chromosome-linked form and an autosomal dominant form of the disease, respectively. Mutations in these genes, however, only account for approximately 20% of all Kallmann syndrome case...
متن کاملIdentification and pharmacological characterization of prokineticin 2 beta as a selective ligand for prokineticin receptor 1.
Prokineticins 1 and 2 (PK1 and PK2) have been recently identified from humans and other mammals and play multiple functional roles. PK proteins are ligands for two G protein-coupled receptors, PK receptor 1 (PKR1) and PK receptor 2 (PKR2). Here, we report the molecular cloning and pharmacological characterization of an alternatively spliced product of the PK2 gene encoding 21 additional amino a...
متن کاملUpdates on Endothelial Functions of Proangiogenic Prokineticin.
Coronary and renal blood flows are important for maintaining cardiac and renal function. Endothelial cells (ECs) play critical roles in forming new vessels and regulating vessel functions. ECs also protect the surrounding tissues by secreting a variety of biologically active substances. Endothelial dysfunction occurs in various forms of cardiovascular and renal diseases. It plays a fundamental ...
